Las propiedades mecánicas anómalas de los tejidos tumorales activan las vías bioquímicas tumorigénicas. Se produce una desestabilización de la homeostasis del tejido adulto en respuesta a tensiones mecánicas patológicas. La hipervascularización en ciertos tumores tardíos genera una presión hidrodinámica estática elevada que puede promover la progresión maligna del tumor a través de la hipoxia o líquido intersticial anómalo y flujo sanguíneo. La alta rigidez de los tumores induce directamente una activación mecánica de vías bioquímicas que mejoran el ciclo celular, la transición epitelial-mesenquimática y la motilidad celular.

Además, los aumentos en la presión del estrés sólido asociado con la hiperproliferación celular activan las vías tumorigénicas en las células epiteliales sanas comprimidas por el tumor vecino. Por ello, comprender estos mecanismos será importante para el desarrollo de tratamientos innovadores para tratar dichas anomalías mecánicas en el cáncer.

Este proyecto presenta dos tecnologías basadas en el uso estratégico de los ultrasonidos de baja intensidad y de la microfluidica para abordar procesos de tumoración, adquirir conocimiento biodinámico sobre el microambiente tumoral y realizar caracterización tenso-elástica en el interior de los tumores. Además, abre una nueva línea de investigación mediante el desarrollo de nuevos sistemas con ultrasonidos para la actuación sobre tumores.

We propose in this project the development of a new non-invasive tool to modulate tumors and stromas based on a low intensity ultrasounds (LIUS) actuation. A suitable modulation of specific physic-chemical parameters is controlled by characterizations of biochemical type (proteins, histology) and acoustical (hardness). It constitutes a very promising ensemble strategy for stopping the advance of very aggressive tumors, as in the pancreas.

In addition, new high-resolution ultrasonic methods will be investigated, for non-invasive early detection of very small physical changes (spatial, thermal and elastic) precursors of a future cancer.
